A noncanonical NF-kB - β-catenin signaling axis in dendritic cells instructs gut inflammation

چکیده
Abstract Dendritic cells (DCs) balance intestinal homeostasis against inflammatory insults. In particular, the noncanonical NF-kB pathway, which activates RelB:p52 heterodimers, has been implicated in DC differentiation. However, if DC-intrinsic signaling modulates function inflamed gut remains unclear. Here we report that genetic ablation of DCs accumulated regulatory T lamina propria and associated lymph nodes, restraining inflammation colitogenic gut. Our mechanistic investigation revealed an impaired gene function, rather unexpectedly, augmented abundance active β-catenin via impairment destruction complex. turn, transcriptionally upregulated Raldh2, imparting a retinoic acid signature tolerogenic functions to NF-kB-deficient DCs. Indeed, mice possessing defects exhibited increased frequency +DCs resilience chemical induced experimental colitis. Noncanonical Wnt5a enhances Wnt/β-catenin signaling during osteoblastogenesis
Wnt regulates bone formation through β-catenin-dependent canonical and -independent noncanonical signaling pathways. However, the cooperation that exists between the two signaling pathways during osteoblastogenesis remains to be elucidated. Here, we showed that the lack of Wnt5a in osteoblast-lineage cells impaired Wnt/β-catenin signaling due to the reduced expression of Lrp5 and Lrp6. Pretreat...
متن کاملMaternal Wnt/β-Catenin Signaling Coactivates Transcription through NF-κB Binding Sites during Xenopus Axis Formation
Maternal Wnt/β-Catenin signaling establishes a program of dorsal-specific gene expression required for axial patterning in Xenopus. We previously reported that a subset of dorsally expressed genes depends not only on Wnt/β-Catenin stimulation, but also on a MyD88-dependent Toll-like receptor/IL1-receptor (TLR/IL1-R) signaling pathway. Here we show that these two signal transduction cascades con...
متن کاملCrosstalk between Wnt/β-Catenin and NF-κB Signaling Pathway during Inflammation
Besides its important role in embryonic development and homeostatic self-renewal in adult tissues, Wnt/β-catenin signaling exerts both anti-inflammatory and proinflammatory functions. This is, at least partially, due to either repressing or enhancing the NF-κB pathway. Similarly, the NF-κB pathway either positively or negatively regulates Wnt/β-catenin signaling. Different components of the two...
متن کاملRe-activation of Wnt/β-catenin Signaling Pathway in Hair Follicle Stem Cells in Treatment of Androgenetic Alopecia
Hair loss is a common hair disorder in human population. It affects quality of life and there are ongoing attempts to find permanent treatment for this condition. But, today there is no completely safe and protective treatment for all. Hair follicle stem cells are alive, but quiescence in androgenetic alopecia and are potentially active and can proliferate and differentiate, then regenerate hai...
متن کاملChemokine GPCR Signaling Inhibits β-Catenin during Zebrafish Axis Formation
Embryonic axis formation in vertebrates is initiated by the establishment of the dorsal Nieuwkoop blastula organizer, marked by the nuclear accumulation of maternal β-catenin, a transcriptional effector of canonical Wnt signaling. Known regulators of axis specification include the canonical Wnt pathway components that positively or negatively affect β-catenin. An involvement of G-protein couple...
